Yes, it works. I've used both hand calls (Faulk's) and my FX3 with excellent results. Sorting through scat will show you that coyotes do indeed eat plenty of squirrels. BTW, I've also called grey fox, bobcat, and about every bird of prey around with the same distressed squirrel sounds.
